p-ERK Antibody (E-4) is an IgG2a κ mouse monoclonal p-ERK antibody (also designated p-ERK antibody or p44/42 MAPK antibody) that detects ERK 1 phosphorylated at Tyr 204 and correspondingly phosphorylated ERK 2 of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA. p-ERK Antibody (E-4) is available as the non-conjugated anti-p-ERK antibody form, as well as multiple conjugated forms of anti-p-ERK antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. The activation of signal transduction pathways by growth factors, hormones and neurotransmitters is mediated through two closely related MAP kinases, p44 and p42, designated extracellular-signal related kinase 1 (ERK 1) and ERK 2, respectively. ERK proteins are regulated by dual phosphorylation at specific tyrosine and threonine sites mapping within a characteristic Thr-Glu-Tyr motif. Phosphorylation at both the Thr and Tyr residues is required for full enzymatic activation. In response to activation, MAP kinases phosphorylate downstream components on serine and threonine. Upstream MAP kinase regulators include MAP kinase kinase (MEK), MEK kinase and Raf-1. The ERK family has three additional members: ERK 3, ERK 5 and ERK 6.

Hello Sir/Mam Should we have to use 5% BSA as the blocking agent, diluent for primary antibody and secondary antibody or is 5% milk okay to use.

Thank you for your question. We recommend using our plant protein-based UltraCruz® Blocking Reagent: sc-516214 for blocking purposes. BSA- or milk powder-based may also be used instead: If signal strength is too low, BSA may be the better choice, whereas a milk powder blocking buffer is better suited to block promiscuous, unspecific signals.

Does sc-7383 recognize dually phosphorylated ERK1/2? What is the biological role of mono-phosphorylated ERK1/2 and does it necessarily undergo threonine phosphorylation subsequently? Thank you.

Thank you for the question. p-ERK Antibody (E-4): sc-7383 is recommended for detection of ERK 1 phosphorylated Tyr at the position 204 with a mw of ~44kDa. Correspondingly phosphorylated ERK 2 will also be detected at ~42kDa. It was not examined if the antibody has a different binding capacity of the dually phosphorylated vs. the mono phosphorylated ERK proteins. ERK proteins are regulated by dual phosphorylation at specific tyrosine and threonine sites mapping within a characteristic Thr-Glu-Tyr motif. Phosphorylation at both the Thr and Tyr residues is required for full enzymatic activation. In response to activation, MAP kinases phosphorylate downstream components on serine and threonine. Yes. We recommend using a directly conjugated version of p-ERK Antibody (E-4) sc-7383. This antibody is currently available conjugated to AlexaFluor 405 (sc-7383 AF405), AlexaFluor 488 (sc-7383 AF488), AlexaFluor 647 (sc-7383 AF647), biotin (sc-7383 B), fluorescein (sc-7383 FITC), HRP (sc-7383 HRP), PCPC5 (sc-7383 PCPC5), phycoerythrin (sc-7383 PE), PerCP (sc-7383 PerCP), and TRITC (sc-7383 TRITC).
