• Mechanism of Action

- Unsiatem® inhibits both the oxidized and reduced form of XO

- Unsiatem® is a non-purine, selective and potent inhibitor of XO, 10-30 times

                         more than allopurinol, and has no effect on other enzymes involved

                         in purine or pyrimidine metabolism

XO = Xanthine Oxidase

XDH = Xanthine Dehydrogenase

- Unsiatem® is a Xanthine Oxidase (XO) inhibitor

Side Effects

The most common Adverse Reactions:

- Liver Function Abnormalities

- Nausea, Arthralgia, Rash and Dizziness

Unsiatem® is contraindicated in patients being treated with azathioprine or

6-mercaptopurine

• Precautions

- After initiation, gout flare frequently occurs;  provide

  prophylaxis with NSAIDs or colchicine upon

  initiation and for ≤6 months

- May increase liver enzyme activity; obtain Liver

  function tests at baseline and do not initiate if AST is

  3x ULN with total bilirubin > 2x ULN
